The polyclonal cell line Jurkat-Luc is a derivative of the wildtype Jurkate E6-1 T-lymphoblast cell line achieved through transduction with a lentiviral expression vector. This vector encodes the codon-optimized firefly luciferase gene (luc2) controlled by the EF-1 alpha promoter, along with the gene for Puromycin N-acetyl-transferase regulated by the hPGK promoter. K562-Luc demonstrates stable expression of luciferase and resistance to Puromycin.

Jurkat-Luc is well suited for quantitative cell viability and proliferation assays as well as cell-based cytotoxicity and immune effector assays, where luciferase signal directly correlates with viable target cell number. In addition, the stable EF-1α-driven luc2 expression enables sensitive bioluminescence-based in vivo imaging for longitudinal monitoring of tumor burden and treatment response.
